Does Disk partitioning do 4K alignment?

I haven't tried the disk configuration feature, and normally edit partitions by hand inside Win7's install setup. When using NTLite's partitioning, does it create a reserved partition (if required) to follow 4K block boundaries? The reason is my target machine's boot disk always requires a 100MB reserved partition to align.

Does NTLite know about 4K alignment, or simply creates a starting partition from 0 MB?

NTLite just creates the file(or however it does it) that tells the os what to do, its the os itself that does the partitioning and formatting, w7 is 4k aware and knows how to format an ssd.

Yes it does, you can see in the UI when switching to UEFI (EFI partition type), it sets the reserved EFI partition size to 260MB, and in the description it warns about reducing it to the optional 100MB if no 4k drives will be used.
You can of course change those sizes manually, even if bigger is needed, this is just a recommended minimum. More info here.

NTLite has many users and thus a lot of feedback like this which makes it mature, keep it coming :)
