I would like to know how to create serial numbers that can only be used once for an iso, with ntlite?

You do not 'create' serial numbers for software unless you have the legal and authorised right to do so.

I hope you mean 'add serial number' to software in the parameters

For OEM systems, there is no need.
BUT
For upgrading OEM systems, make a batch that reads the serial number and install the specific license key to that system's serial number.
Call or integrate the batch in Post-setup page, in the lower pane.
